Mesenchymal stem cells (MSCs) extracellular vesicle production serum-free medium is a specially designed extracellular vesicle production medium by Tongteng Xinchuang for human mesenchymal stem cells (including umbilical cord mesenchymal, bone marrow mesenchymal, adipose derived mesenchymal stem cells, etc.), suitable for use in the MSC extracellular vesicle production harvest stage. A certain proportion of nutritional additives can also be added to prepare a complete culture medium for use in the cell expansion and harvesting stages of MSC extracellular vesicle production.

Mesenchymal stem cell nutritional supplement is a human platelet lysate (hPL), which is non heterologous and does not contain animal serum. It is mainly used as a substitute for animal serum in traditional culture media. Nutritional supplements contain abundant growth factors and cytokines, aimed at providing the necessary nutrients for cell growth and proliferation.

Mesenchymal stem cell extracellular vesicle production serum-free medium is a medium used to increase the production of extracellular vesicles in stem cells. It can support 2D and 3D cell culture modes, and compared to similar domestic media, the extracellular vesicle production can be increased by more than 50%. And it can greatly reduce batch differences, lower the risk of contamination from various viruses, mycoplasma, etc

This set includes 2L (500mL × 4) serum-free culture medium for the production of extracellular vesicles from mesenchymal stem cells, as well as 100mL of mesenchymal stem cell nutritional supplements.


Complete preparation of culture medium


1. Before use, thaw the mesenchymal stem cell nutritional supplement in a 37 ℃ water bath.

2. Add 5% mesenchymal stem cell nutrient additive (i.e. 25ml nutrient additive to 500ml culture medium) to serum-free medium for the production of extracellular vesicles from mesenchymal stem cells, and prepare it into a complete culture medium before use.

3. Please use the prepared complete culture medium immediately or store it at 2-8 ℃ and use it up within 2 weeks.


Extracellular vesicle production

1. Cell recovery and seed expansion: After cell recovery, use the original complete culture medium (recommended to use the mesenchymal stem cell serum-free basic culture medium set, item number: C04.004.003) for culture and expansion until the exosome production stage;

2. Inoculate and amplify extracellular vesicle producing cells using the complete culture medium prepared with this product (recommended to use a CEL-G® Culture Ad60 fixed bed bioreactor).

3. During the harvest stage of MSC extracellular vesicle production, which is 3-4 days after the cells have grown in complete culture medium, it is possible to independently choose whether to add nutritional additives to the culture medium according to process requirements.

4. If you choose to use complete culture medium, you can directly harvest the supernatant.

5. If choosing not to add nutritional supplements, rinse the cells with 1X PBS solution;

6. Then add mesenchymal stem cell exosomes to produce serum-free culture medium and continue culturing for a period of time. Collect the cell culture supernatant, which can be used for exosome extraction.



Recommended Use
